Lateral spatial assay of zinc and superoxide dismutase in the ocular lens.

چکیده
A procedure is described, using a freezing microtome and measurements of tissue slice volume, to obtain serial sections of the lens for analysis of components from one side of the organ to the other. The concentrations of zinc (analyzed by atomic absorption spectrophotometry) were similar in serial sections taken across the lens from the anterior to the posterior surface. The levels of the zinc metalloenzyme superoxide dismutase were, however, much higher in the anterior sections. This quite simple procedure may be useful for analyzing the distribution of lenticular components.

Superoxide dismutase isoenzymes in the human eye.
PURPOSE The authors determined the levels of the secreted, interstitially located extracellular superoxide dismutase (EC-SOD), the cytosolic copper- and zinc-containing SOD (CuZn-SOD), and the mitochondrial manganese-containing SOD (Mn-SOD) in tears, cornea, sclera, aqueous humor, lens, vitreous body, and retina of the human eye. METHODS SOD enzymatic activity was determined by the direct spe...

Background & Objective: The purpose of study, was to investigate the effect of 8weeks incremental aerobic training with zinc supplementation on muscular enzymatic activity of superoxide dismutase and serum leptine in wistar rats. Materials & Methods: 32 Wistar rats, randomly divided into 4groups (control, training, training +zinc and zinc). The training protocol consisted of running on treadmil...
متن کاملFluorescence assay for monitoring Zn-deficient superoxide dismutase in vitro.
A method has been developed for selective detection of the zinc-deficient form of Cu, Zn superoxide dismutase (SOD1) in vitro. Zinc-deficient SOD1 mutants have been implicated in the death of motor neurons leading in amyotrophic lateral sclerosis (ALS or Lou Gerhig's disease). Thus, this method may have applicability for detecting zinc-deficient SOD1 mutants in human ALS patients samples as wel...
متن کاملDifferent immunoreactivity against monoclonal antibodies between wild-type and mutant copper/zinc superoxide dismutase linked to amyotrophic lateral sclerosis.
Although more than 100 mutations have been identified in the copper/zinc superoxide dismutase (Cu/Zn-SOD) in familial amyotrophic lateral sclerosis (FALS), the mechanism responsible for FALS remains unclear. The finding of the present study shows that FALS-causing mutant Cu/Zn-SOD proteins (FALS mutant SODs), but not wild-type SOD, are barely detected by three monoclonal antibodies (mAbs) in We...
متن کاملSuperoxide Dismutase from Lens esculenta: Purification and Properties.
Superoxide dismutase has been purified to homogeneity from Lens esculenta cotyledons and shoots. The two forms appeared to be identical. The purified enzyme contained two electrophoretically distinct bands. It contained two ions of Cu and two ions of Zn. Gel filtration experiments indicate a molecular weight of about 33,000. The spectrum of ultraviolet and visible regions and electron paramagne...
